1: Roles
Obviously, players should play as themselves. However, you should randomly assign a few of your players secret agendas/information. You can text each player their information, or write them note cards. Possible notes could include:
Unless you're willing to break a whole bunch of laws, there's no getting around the lack of actual death. However, by giving every player a unique role, you can more strongly drag them into the world.
Additionally! You get the added benefit of players not being able to fully trust each other. Just don't add too many 'evil' roles, and it can add an extra layer.
2: Set-Up
I've read enough of your comments to know that this is a low budget project. With that in mind, you can still get a lot done with a small amount of cash. It's all in how you spend it.

These could be used for puzzles mostly. Assuming that everyone is playing by the rules, you shouldn't need to actually lock the doors. At best, you could mount a calculator next to each one. That would allow people to type in their numbers, and add them up. That way, you won't end up with any accidental cheaters.
You can place files with relevant info in puzzle rooms. By doing this, you could make morse code, binary, or similar puzzles far more fair. Most people don't know codes of those varieties off of the top of their heads, so this would be a fair way to implement them.
For the AB Game, if you choose to do so, the only thing you'd NEED to do is separate players. After that, you can handle it any number of ways. They could write their results on note cards, text you their answers, or just say them to you.
There are a lot of good suggestions for puzzles in this thread, so I'll leave that to others.
3: Gonna be honest, I don't have the time to really add anything else to this. Like, maybe I'll edit some more in later, but probably not.
