Um, sort of. Wordpress itself needs to be run on a server, so you can't work with it offline unless you download a development server like easyphp.
You can design the website in just HTML and CSS on your computer, but then you'll have to make significant changes when it's time to put it into WordPress. Instead of HTML files, WordPress uses PHP files (with HTML content in them). I usually just go straight from design to WordPress template files, and skip the HTML mockup part.
It's actually not that easy to jump straight into WordPress without any HTML or CSS experience, but it can be done. Here are some books you can check out to get you started:
