Creative teams change often and characters get re-worked e.g. I never cared for Hawkeye until Matt Fraction’s run. Focus on well-received and relatively self-contained stories. Pick an interesting character/team and seek their “greatest hits”. You may encounter the occasional unexplained reference/character/event—just ride along (Wiki if necessary).

The Defenders are actually very, very different in the comics, Jessica Jones isn't even a member of the team. Although all of the characters are very close friends in the comics and appear in each others series fairly often (when Jessica, Luke, and Danny have their own series that is). I know that Luke Cage, Jessica Jones, and Daredevil are members of the Mighty Avengers, but have no idea if Iron Fist is also a member (the Avengers history and line-up isn't really my area of expertise). When it gets closer to the release of The Defenders on Netflix, however, I'd almost put money on Marvel restarting the Defenders with the line-up of the series, so look out for that.

I would encourage you not to worry so much about what's canon and what isn't. Ninety-nine percent of what Marvel publishes is canon, and non-canon stuff is almost always set in the future or something like that, so it's easily identifiable.
For the characters you're interested in, here's some good starting points I recommend:
Make sure to check your local library first, they may have a lot of these available for you to check out. I'd also want to let you know that Marvel has a digital subscription service called Marvel Unlimited that has almost everything I'm recommending available to read for just $10/month or $70/year. It's a real good deal.

Remember, there are many great characters, creators, publishers, etc. to explore.
Pick an interesting character/team and seek their most popular/acclaimed stories. Focus on self-contained, complete stories in one corner of the universe. There will be unexplained references/characters, just persevere or Wiki. Don’t let the tangled web of shared-universe comics overwhelm you. Think of it like solving a jigsaw puzzle one small piece at a time until you finally see the big picture.
Discover your preferences and let them guide you. Don’t get stuck preparing/over-analyzing, just start reading. Do you like/dislike old/new comics? Specific writers/genres? Cartoony/realistic art?
