Similar to the Vanguard 49 and the Tamrac Evolution mentioned by others, I use one with a horrible name: QuantaRay Urban Gear U100 Pro Backpack, for about $90 new at various camera stores.

There aren't many reviews on it, but I found some images of it showing it off. It is actually multiple parts and a little hard to describe. Look over the pictures with description.

Since there are no good reviews, I'll describe it.

First is the backpack shell. It is fairly hard, like a luggage. It has a full 180-flip back panel. Inside the backpack is a completely removable second camera carrier I'll cover second. Outside the back panel are two pockets that run down to the base. Inside it is it is padded, with multiple pockets on the inside the outermost panel. The strap side of the backpack has a second compartment double-padded against the wearer's back, which fits a slender notebook or tablet, papers, binder, or similar items. It comfortably holds my laptop (14" screen), but if you have a monster laptop it won't fit. It short external straps I use for my tripod, and is rugged and waterproof.

So that's the big hard backpack. Very padded, rugged, has a huge main body, a smaller second cavity for the laptop, and a few pockets, and tripod cables.

Inside the backpack is a completely removable camera case, the same size as the backpack. It also opens fully for a 180-flip, is waterproof, and fairly rugged. The outside has handles for carrying. Inside it are a clear zippered pouch, a very large open pocket, and an open area with a bunch of padded velcro-attached pieces that you can adjust for your needs. It is about 7 inches deep, I keep my lenses standing upright. My bag is rather full with 4 lenses, 2 flashes, pocket wizards, about 10 cokin filters and 2 filter holders plus some rings, the camera with a lens attached, and some assorted trappings like batteries, cables, wipes, etc.

The only downside is the gear is secured enough that I need to take it off to reach inside, but really, I wouldn't want to be wearing it and accidentally dump everything, so that's a good thing. Again, super rugged with multiple waterproof layers.
