Okay, Lets break this down.
If you are trying to wake up the Windows PC from external computer you will need to configure the Router. Otherwise you should not, for this example I will skip this example as it sounds like you are doing everything from the same local network.

I don't use Amazon Firestick (You mean Amazon Fire TV Stick?? ;)) .
I found this app (https://www.amazon.com/J%C3%B6rn-Buitink-Wake-On-LAN/dp/B00Q581LPO) . It looks like you will need to set info for you PC up on your Fire TV Stick.
You will need to configure the app on your Fire TV Stick. With the following Info:

MAC Address: Example: 0W:00:X0:WE:04:08

IP Address: Example: 192.168.1.70

Port # : Example: 9


You can then use this app to start up your PC, and then once it boots up it will start plex. You can switch to the plex app.
