I'm sure you could do it without the adjuster tool but the tool makes it so incredibly easy. Here's a kit with everything you need for $50:

Also, the vacuum test ports are on the intake runners, not the carbs.

Drain fuel from your float bowls into a glass jar and look for water, if more than a bowls worth comes out and it keeps flowing, you know the petcock and filters are OK...then a spray of starting fluid into the air box while starting tells you if you have spark, though it doesn’t check all 4 plugs like removing them does. Just trying to suggest the least intrusive way to check those items you mentioned. You may need to get a 4-carb synchronizer, maybe a shop will rent one. Like this one.
