Not hard if you can keep things square and plumb. Read up on king studs, door/window headers and fire stops before you begin.
Cost is harder to figure as you didn't give dimensions. But figure 2 2x4 boards for the whole length of the wall, a 2x6 twice the length of the width of the door (plus a scrap of plywood the same length x 6" that you may be able to get free out of their cutting scraps), and then a 2x4 for every 16" of your wall. Add all that up and multiply by your local wood costs.
edit: Oh yeah..and if you have a cement floor in the garage, look into renting/borrowing one of these or a similar product.
